Parker, 22, dominated his 39-year-old opponent on Saturday, winning via a seventh-round technical knock out.
The Aucklander knocked the much Minto to the canvas in the fifth round with a right hook and the much smaller American took more blows in the seventh round, prompting his management to throw in the towel.
Fighting at a reduced weight of 102kg, Parker used his speed and jab to dominate the early rounds.
He didn't allow his opponent to come inside for any close exchanges, as Minto had done so effectively in beating another big-name Kiwi fighter, Shane Cameron, in December.
"I executed the plan we put in place," Parker said.
"I told everyone I was going to be lean and mean. We've developed speed and power but we're still at the beginning stages."
Minto, who suffered an eighth loss in 38 fights, cedes his WBO Oriental championship title to Parker.
Earlier, leading New Zealand light heavyweight Robert Berridge delivered a first round knockout of Brazilian Rogerio Damasco.
It was a 20th knockout win for Berridge (24-1-1), who retained his twin WBO regional belts and added weight to the 29-year-old's claim for a title fight against Russian WBO champion Sergey Kovalev.
Unbeaten Australian welterweight Jeff Horn, 26, underlined his potential with a dominant 120-107 points win over Brazilian Fernando Ferreira de Silva.
The Brisbane schoolteacher overcame the pain of a broken hand suffered in the sixth of 12 rounds to notch an eighth win and defend his WBO Oriental title.
